New Records of Xylaria Species in Korea: X. ripicola sp. nov. and X. tentaculata

Abstract: During a Korean mushroom diversity survey from 2011 to 2014, we found one new Xylaria species (X. ripicola sp. nov.) and one Xylaria species that had not been previously observed in Korea (X. tentaculata). To confirm the phylogenetic placement of the new species, we conducted a phylogenetic investigation based on internal transcribed spacer regions of ribosomal DNA sequences. Additionally, the new species, X. ripicola, was subsequently analyzed for RNA polymerase II subunit sequences. We also evaluated the mac… Show more

“…The molecular phylogeny was inferred from a combined dataset of ITS, TUB and RPB2 sequences. PCR reactions were performed in a total volume of 20 µl consists of 6 µl distilled water, 10 µl 2x PCR master mix (iNtRONbiotechnology, Korea), 1 µl of each 10 µM primer (0.5 µM) and 2 µl template DNA.…”

“…It is di cult to differentiate closely related species. However, molecular techniques such as polymerase chain reaction (PCR) for mushroom identi cation have proved to be fast, speci c, and sensitive, especially, inconsistent morphology and indiscrimination among species of mushrooms (Cho et al 2015;Kim et al 2016;Lee et al 2000;Persoh et al 2009;Raja et al 2017). The conserved region such as Internal Transcribed Spacer (ITS) of nuclear ribosomal DNA (rDNA) usually has been targeted because it is highly conserved among interspecies but variable between intraspecies (Raja et al 2017).…”

“…The Internal Transcribed Spacer (ITS) of nuclear ribosomal DNA (rDNA) is the conserved region and usually has been targeted for fungal identification due to being highly conserved inside the same species and variable among species in this region. (Kim et al, 2016;Raja et al, 2017). However, in several fungal taxa, molecular identification using ITS alone could not be certain.…”
