New Small Plasmid Harboring bla KPC-2 in Pseudomonas aeruginosa

Abstract: The aim of this study was to characterize the genetic context of bla KPC-2 in Pseudomonas aeruginosa sequence type 244 from Brazil. The bla KPC-2 gene was detected in a new small plasmid, pBH6. Complete sequencing revealed that pBH6 was 3,652 bp long and included the Tn3 resolvase and Tn3 inverted repeat (IR), a partial copy of ISKpn6, and a putative ori region but no rep genes. pBH6 replicated stably into Escherichia coli strain DH10B and P. aeruginosa strain PAO.

“…Interestingly, large direct repeats could flank genomic rearrangements between NTE KPC elements and small mobilizable plasmids. In fact, recent studies have reported the presence of these small plasmids in KPC-2-producing Pseudomonas aeruginosa and Escherichia coli and in BKC-positive Klebsiella pneumoniae isolates (12)(13)(14)(15).…”

“… 4 Up to now, five fully sequenced bla KPC -carrying plasmids from P. aeruginosa are available in public databases: two IncP-6 plasmids pCOL-1 (accession number KC609323) and p10265-KPC (accession number KU578314) from Colombia 5 and China, 6 respectively, two IncU plasmids pPA-2 (accession number KC609322) and pD5170990 (accession number KX169264) from Colombia 5 and Brazil, respectively, and pBH6 (accession number CM003767; it could not be assigned into any of known incompatibility groups) from Brazil. 7 Each of pCOL-1, p10265-KPC, pPA-2, and pBH6 contains the sole resistance gene bla KPC-2 , while pD5170990 carries bla KPC-2 , strAB (aminoglycoside resistance), cmx (chloramphicol resistance), qacED1 (quaternary ammonium compound resistance), and sul1 (sulphonamide resistance).…”

“…Furthermore, it was also noted that the BH9 Mu-like prophage was also inserted on the P. aeruginosa BH9 chromosome, exhibiting a distinct 5-bp direct repeats (TCTGC), but not containing the pBH6 sequence and the bla KPC-2 gene ( Figure 1 ). The presence of both, BH9 Mu-like prophages, strongly indicates for a previous event of mobilization of the BH9 Mu-like phage by its replicative transposition mechanism, directly molding the P. aeruginosa BH9 genome and impacting the bla KPC-2 mechanism of propagation by the small plasmid pBH6 ( Galetti et al, 2016 ).…”

“…It has previously been reported that the bla KPC-2 gene in P. aeruginosa BH6, which was isolated from the same nosocomial environment, was harbored by a small plasmid pBH6 (3,652 kb) ( Galetti et al, 2016 ). Indeed, comparative genomic analysis of the assembled P. aeruginosa BH9 genome revealed that the bla KPC-2 gene was included in an additional and larger contig containing identical sequence to the pBH6 plasmid, and also encoding several additional phage-related genes (hereafter called prophage BH9).…”
