Nicotinic Receptors Underlying Nicotine Dependence: Evidence from Transgenic Mouse Models

Abstract: Nicotine underlies the reinforcing properties of tobacco cigarettes and e-cigarettes. After inhalation and absorption, nicotine binds to various nicotinic acetylcholine receptor (nAChR) subtypes localized on the pre-and post-synaptic membranes of cells, which subsequently leads to the modulation of cellular function and neurotransmitter signaling. In this chapter, we begin by briefly reviewing the current understanding of nicotine's actions on nAChRs and highlight considerations regarding nAChR subtype localiz… Show more

“…These receptors can be activated by both endogenous ligand, ACh, and exogenous ligands, such as nicotine. ,, They are also involved in the regulation of processes like cell excitability, transmitter release, and neuronal integration by acetylcholine-mediated innervation, which contributes to their critical role in modulating a wide range of physiological processes, including pain, fatigue, cognition function, sleep, and arousal . nAChRs are critical in early pre- and perinatal circuit formation and age-related cell degeneration, making them an important target in the pathogenesis of various neurological disorders and neurodegenerative diseases such as AD, PD, myasthenia gravis, and atherosclerosis. , …”

“…Nicotine acts on ionotropic nicotinic acetylcholine receptors (nAChRs), where ligand binding results in channel pore opening and cation influx across the membrane ( 89 ). nAChRs are pentameric, assembling with various combinations of α and β nAChR subunits for a wide variety of nAChR subtypes with distinct pharmacokinetics, expression patterns, and actions on the cholinergic system ( 90 , 91 ). Acting on nAChRs, nicotine exerts its reinforcing properties through the mesolimbic dopaminergic pathway ( 92 ).…”

“…Excitation of the vagus nerve (VN) in the lung model causes cell proliferation in lung cancer via nicotine acetylcholine receptors (nAChRs). The proinflammatoryinduced microorganisms play this role by activating TLR-4 expression and signaling VN to mediate inflammation through bacterium products [30], [31]. Nicotine and acetylcholine activate nAChR are found in neurons and non-neuronal cells.…”
