NIR-II Responsive Nanohybrids Incorporating Thermosensitive Hydrogel as Sprayable Dressing for Multidrug-Resistant-Bacteria Infected Wound Management

Abstract: Developing an effective dressing against bacterial infection and synchronously addressing wound complications, such as bleeding, long-term inflammation, and reinfection, are highly desirable in clinical practice. In this work, a second near-infrared (NIR-II) responsive nanohybrid consisting of imipenem encapsulated liposome with gold-shell and lipopolysaccharide (LPS)-targeting aptamer, namely ILGA, is constructed for bacteria elimination. Benefiting from the delicate structure, ILGA exhibits strong affinity a… Show more

“…It should be noted that the proportion of CD206+ macrophage in LPS treatment group is higher than that of PBS group (Figure C). This can be reasoned that the macrophage population in the LPS-induced inflammatory microenvironment stimulates a small fraction of macrophages to polarize toward the M2 phenotype to resist excessive oxidative stress and inflammatory mediators for the purpose of survival, in agreement with the literature. , In any case, this is not contradictory to the claim that LPS treatment mediates M1 polarization of macrophages since the overall ratio of CD206/CD86 (M2/M1 macrophage ratio) after LPS treatment is lower in the LPS group than in the PBS group, meaning that the overall macrophages have been polarized to the M1 phenotype (Figure D).…”

“…49 Flow cytometry is further used to evaluate the macrophage phenotype. 50 Compared with H and PBS, the RAW 264.7 cells in H-Fe 2+ -Zn 2+ , H-Fe 2+ -Zn 2+ -NIR, and H-Zn 2+ had a progressive tendency to upregulate CD206, ranging from 14.7% to 18.7%, until 19.2% (Fig. 4a, b and Fig.…”

“…The sol–gel-precipitation phase transition temperature curves of copolymer solutions with varying concentrations were determined by using the test tube inversion method (with 1 °C temperature intervals). The lower critical gelation temperature (LCGT) was defined as the temperature at which the sol–gel transition occurred, while the upper critical gelation temperature (UCGT) was defined as the temperature at which gel-precipitation transition occurred . Rheological study was performed using a Discovery Hybrid Rheometer 2 (TA Instruments, USA).…”
