NMR spectroscopic identification of cholesterol esters, plasmalogen and phenolic glycolipids as fingerprint markers of human intracranial tuberculomas

Abstract: Detailed (1)H and (13)C NMR spectroscopy of lipid extracts from 12 human intracranial tuberculomas and two control brain tissue samples was performed to assess the role of lipids in the disease process. One-dimensional and two-dimensional NMR techniques were used to resolve the mixture of lipid components and make resonance assignments. The lipid components that could be identified in tuberculoma lipid extracts and not in control samples were: cholesterol ester, plasmalogen and phenolic glycolipids. It is prop… Show more

“…Hence the chains are not only slowly diffusing (as is apparent from diffusion edited spectra) but also free to rotate. This situation is usually indicative of the presence of lipid bodies (LB) in the cell [24,25,27]. As reported above, the NMR data show that these lipids are essentially constituted by mobile TAG.…”

“…In the spectra of n-3 LC-PUFA-supplemented cardiomyocytes it is possible to recognize the characteristic signals from unsaturated FA. The resonances of FA chains are identified as: 1, methyls (-CH 3 ) at 0.89 ppm (saturated FA chains), and 2, -CH 3 at 0.96 ppm (n-3 EPA, and DHA); 3, acyl chain methylenes (CH 2 ) n (1.33 ppm); 4, CH 2 -C-CO (1.58 ppm); 5, CH 2 C=C (2.02 ppm); 6, CH 2 CO other than DHA (2.25 ppm) and 7, CH 2 CO of DHA, (2.33 ppm); 8, =C-CH 2 -C= (2.78 ppm); 9, -CH=CH-(5.33 ppm) [24][25][26]. Signals at 2.02 ppm are assigned to methylene protons of the CH 2 -CH=CH moiety of both monounsaturated FA (MUFA) and PUFA, and the signal at 2.78 ppm is characteristic of PUFA [24,25].…”

“…The assignments of adenosine-3-phosphate were confirmed by comparison with the spectra of the same compound downloaded from BMRB [31]. The broad resonances of macromolecules and fatty acids in 1 H spectra were assigned using the data in the literature [32][33][34][35] In the 31 P spectra, once assigned GPC signal at 0.48 ppm, the pH was calculated using the chemical shift distance between the Pi signal and the GPC peak [14,16]. According to this, the calculated pH of each sample is shown in the Table S1 (see Electronic Supplementary Material).…”
