No deleterious effect of low dose methotrexate on titanium implant osseointegration in a rabbit model

Abstract: OBJECTIVE:To evaluate the effect of low dose methotrexate alone or in combination with glucocorticoid treatment on titanium implant osseointegration.METHODS:Groups of 6–8 adult New Zealand White rabbits were treated for 18 weeks with saline (control), methotrexate, glucocorticoid, or methotrexate plus glucocorticoid. The animals received a titanium implant in the tibia at week 6. Lumbar spine and tibia bone mineral densities were analyzed before and after treatment. Histomorphometric analysis of bone cortical … Show more

“…Carvas et al . [ 16 ] evaluated the osseointegratiom process in a rabbit model during an 18-week period. Groups of 6-8 rabbits were treated with saline, methotrexate, glucocorticoid, or methotrexate plus glucocorticoid and received implants after 6 weeks.…”

“…In a study using a canine model, a low-dose MTX reduced bone-to-implant contact (BIC), although osseointegration of inserted implant was acceptable (Tavakoli et al, 2018). On the other hand, another study demonstrated that cortical thickness, total bone area and BIC were not significantly different between the control and MTX groups in a rabbit model (Carvas et al, 2011). In a retrospective case series analyzing implant treatment in patients with RA and connective tissue disease (CTD), 13 implants were inserted in patients receiving MTX, and they all survived (Weinlander et al, 2010) (Table 4).…”

“…Such a medication cocktail may prevent osseointegration and bone repair processes. 334 There is no significant risk factor for implant osseointegration in RA patients despite the apparent ability of RA medications to interfere with it. 335 Consequently, dental implants are not contraindicated by RA.…”
