Nobel somatosensations and pain

Abstract: The Nobel prices 2021 for Physiology and Medicine have been awarded to David Julius and Ardem Patapoutian "for their discoveries of receptors for temperature and touch", TRPV1 and PIEZO1/2. The present review tells the past history of the capsaicin receptor, covers further selected TRP channels, TRPA1 in particular, and deals with mechanosensitivity in general and mechanical hyperalgesia in particular. Other achievements of the laureates and translational aspects of their work are shortly treated.

“…For example, mechanical stimulus is a common interoceptive signal in many organs. Interestingly, although at the macroscopic level mechanical stimuli may trigger distinct forms of tissue deformation in different organs, a family of mechanosensitive ion channels, PIEZO1 and PIEZO2, is widely employed to sense such mechanical changes all over the body ( 5 ). This seemingly surprising observation that one class of receptor could function under so many conditions is likely because only microscopic changes such as physical displacement of molecules and changes in cell membrane curvature can be directly sensed at the molecular level ( 6 ).…”

“…TRPV1 is a non-selective cation channel with relatively high permeability to Ca 2+ ions, which is a major player in pain perception activated by different factors, including heat, inflammation, and acidic environment as revealed by David Julius, that shared the 2021 Nobel Prize for Physiology or Medicine [158][159][160]. Indeed, TRPV1 plays a key role in acidosis-induced pain, acting as a proton channel and being directly activated by protons [78,161].…”
