Non-Coding RNAs Regulate the Resistance to Anti-EGFR Therapy in Colorectal Cancer

Abstract: Colorectal cancer (CRC) is the third prevalent cancer worldwide, the morbidity and mortality of which have been increasing in recent years. As molecular targeting agents, anti-epidermal growth factor receptor (EGFR) monoclonal antibodies (McAbs) have significantly increased the progression-free survival (PFS) and overall survival (OS) of metastatic CRC (mCRC) patients. Nevertheless, most patients are eventually resistant to anti-EGFR McAbs. With the intensive study of the mechanism of anti-EGFR drug resistance… Show more

“…For example, some lncRNAs can be processed to generate miRNAs that negatively regulate target mRNAs [ 41 ], and lncRNAs and circRNAs can act as a ceRNA to competitively bind to miRNAs and eliminate the inhibition of target genes [ 29 , 31 ]. Chu et al have reviewed the advance of ncRNAs in anti-EGFR mAbs therapy resistance [ 190 ]. In this paper, we conducted a more comprehensive search and update of relevant literature, which not only included more reports of ncRNAs related to anti-EGFR resistance, but also demonstrated the interaction networks between different ncRNAs, providing more detailed and in-depth mechanism exploration and researches on reversal of Cetuximab resistance.…”

“…Moreover, the high expression of exosomal miR-92a-3p in serum predicts metastasis and chemotherapy resistance in CRC patients[ 24 ]. The overexpression of miR-100, miR-125b, miR-27a, miR-135, and MIR100HG in CRC tumors modulates the canonical Wnt pathway, promoting events such as proliferation and invasion[ 71 , 72 ].…”

“…Two years earlier, Fang and his group revealed that circ_100290 promotes CRC progression through activation of the Wnt/β-catenin pathway[ 74 ]. Other cRNAs involved in the activation of this signaling pathway in CRC are Hsa_circ_0005075[ 75 ], circMTO1[ 76 ] and Circ-PRKDC[ 72 ]. On the other hand, cRNAs such as circPRKDC and cirITCH act by inhibiting the activity of the β-catenin pathway[ 77 , 78 ].…”

“…Recent studies have revealed that abnormal expressions of long non-coding RNAs (lncRNAs), which are RNAs longer than 200 nucleotides and without the ability to code for proteins, are linked to the occurrence, progression, recurrence, metastasis, and chemoresistance of CRC, which can assist in diagnosis, determining prognosis, and providing therapeutic targets in CRC [ 7 , 8 , 9 , 10 ]. The lncRNAs are often expressed at lower levels but show more cell type-specific expression patterns than protein-coding genes [ 11 ].…”
