Non-HLA-Type Endothelial Cell Reactive Alloantibodies in Pre-Transplant Sera of Kidney Recipients Trigger Apoptosis

Abstract: The vascular endothelium of transplanted organs represents an important target for allograft-directed immune responses. Although HLA antigens expressed on graft endothelial cells (EC) can become targets of the host immune response, the role of other, non-HLA-encoded EC antigens has been proposed but is still unclear. The aim of this study was to investigate the presence of and to characterize anti-EC antibodies (AECA) in 57 kidney transplant recipients according to their HLAimmunization status. Flow cytometry … Show more

“…Endothelial Cell Culture and Activation-Human ECs were isolated from human umbilical veins (HUVECs) or aorta (HAECs) as previously described (20). ECs were cultured in endothelial cell growth medium supplemented with 10% fetal calf serum (FCS), endothelial cell growth supplement (0.4%), hydrocortisone (1 g/ml), human basic fibroblast growth factor (1 ng/ml), human epidermal growth factor (0.1 ng/ml), amphotericin B (50 ng/ml), and gentamicin (50 g/ml) (Promocell, Heidelberg, Germany) at 37°C in a 5% CO 2 humidified air incubator.…”
