Non-invasive in vivo molecular imaging of intra-articularly transplanted immortalized bone marrow stem cells for osteoarthritis treatment

Abstract: Pathophysiology of osteoarthritis (OA) is characterized by progressive loss of articular cartilage in the knee-joints. To impart regenerative ability in lowly metabolizing chondrocytes, the bone marrow stem cells (BMSCs) has recently been recognized as a superior alternative treatment for OA. However, study of primary BMSCs-mediated chondrogenesis is difficult due to progressive cellular aging and replicative senescence. To obtain a therapeutic cell population for OA, BMSCs were immortalized by human papilloma… Show more

“…It is well known that KOA is distinguished by the erosion of articular cartilage, which possesses poor self-healing capacity due to the limited regeneration in chondrocytes [ 41 ]. Our staining and Western blot results showing degraded cartilage and proteoglycans agree with osteoarthritic characteristics.…”

“…Luciferase reporters can also be used in transplanted cell tracking experiments using the same optical imaging systems to demonstrate the presence of exogenous cells in repairing tissues of OA rats following ACLT 55,56 and persistence of transplanted senescent auricular chondrocytes when injected into mouse joints to cause OA like disease 57 .…”

“…MSCs are capable to differentiate into osteoblasts and also secrete factors such as VEGF and BMP-4 to promote bone cell regeneration [ 167 , 168 ]. Studies have also used immortalized BMSCs in osteoarthritic recovery [ 169 ]. These studies showed the potential of MSC therapy in bone-associated disorders.…”
