“…After that it becomes saturated for both the normal cells and the NSCLC cells. The ΔI saturation indicates that 24 h is needed for a cell to completely adhere to the PSW surface, which is consistent with the data reported in (Baumann et al 1999) (Lehmann et al 2000) (Otto et al 2003) (Cai et al 2009). The other item we observed from Fig.…”

“…Generally, it takes 1~2 days for a cell to completely adhere to a substrate (Baumann et al 1999) (Lehmann et al 2000) (Otto et al 2003) (Yu et al 2009). We therefore measured the current changes ΔI of the PSW at V DS =5V for the single normal cells and the NSCLC cells from the very beginning of seeding the single cell into the isolation window as a function of time.…”

“…The sensor chip enables continuous measurement of oxygen consumption using oxygen-sensitive electrodes [27], pH changes of the medium by employing ionsensitive field-effect transistors [28], and the impedance between two interdigitated electrode structures [29] to register the impedance under and across the cell layer on the chip surface. Before measurement, cells were seeded on the sensor chip (SC 1000) in DMEM (PAA, E15-883) with penicillin/streptomycin and 10% (v/v) FCS (PAA) and incubated in a standard tissue culture incubator at 37°C/5% CO 2 and 95% humidity for 24 h until 80-90% confluency was reached.…”

“…The impact of complexes 1 and 2 and 11 and 12 on the metabolism of adherent MCF-7 cells was monitored using a Bionas biosensor chip system, whose silicon chip contains two interdigitated electrode structures that register the cellular impedance [29], miniature Clarke-type oxygen electrodes for recording the cellular oxygen uptake [27], and ion-sensitive field-effect transistors to monitor pH changes of the medium [28]. As the cells were plated at high density in the Bionas system, a much lower proliferation rate was observed than in the crystal violet assay, where MCF-7 cell duplication took about 30 h. The impedance value reflects the integrity of the cellular layer but not the total number of cells and its decrease therefore provides a measure of the extent of cell death rather than the level of antiproliferative activity.…”

“…It is illuminating to contrast the previous application with another physiological application of pH ISFET, in which changes of local pH have been measured with a wellpassivated transistor [9]. In this case the signal provided true chemical information.…”
