The relationship among the gut microbiome, global fecal metabolites and rheumatoid arthritis (RA) has not been systematically evaluated. In this study, we performed 16S rDNA sequencing and liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based nontargeted metabolomic profiling on feces of 26 untreated RA patients and 26 healthy controls. Twenty-six genera and forty-one MS2-identified metabolites were significantly altered in the RA patients. Klebsiella, Escherichia, Eisenbergiella and Flavobacterium were more abundant in the RA patients, while Fusicatenibacter, Megamonas and Enterococcus were more abundant in the healthy controls. Function prediction analysis demonstrated that the biosynthesis pathways of amino acids, such as L-arginine and aromatic amino acids, were depleted in the RA group. In the metabolome results, fecal metabolites including glycerophospholipids (PC(18:3(9Z,12Z,15Z)/16:1(9Z)), lysoPE 19:1, lysoPE 18:0, lysoPC(18:0/0:0)), sphingolipids (Cer(d18:0/16:0), Cer(d18:0/12:0), Cer(d18:0/14:0)), kynurenic acid, xanthurenic acid and 3-hydroxyanthranilic acid were remarkably altered between the RA patients and healthy controls. Dysregulation of pathways, such as tryptophan metabolism, alpha-linolenic acid metabolism and glycerophospholipid metabolism, may contribute to the development of RA. Additionally, we revealed that the gut microbiome and metabolites were interrelated in the RA patients, while Escherichia was the core genus. By depicting the overall landscape of the intestinal microbiome and metabolome in RA patients, our study could provide possible novel research directions regarding RA pathogenesis and targeted therapy.

Background As a Traditional Chinese Medicine with an antipyretic effect, Indigo Naturalis (IN) has a long history of application in China. It is often used with other heat-clearing drugs to treat high fever in children, which is different from other heat-clearing medicines. But its antipyretic effect and mechanism have not been fully elucidated yet. And the heat-clearing development of IN lacks convincing current evidence. Methods This study aimed to uncover the antipyretic effect and mechanism of IN on infectious fever by rectal temperature, pyrogen cytokines and metabolomics. Materials and Methods Firstly, the antipyretic effect of IN on LPS-induced fever rats was evaluated by the changes in rat rectal temperature. Then, the content of PGE2 and cAMP in the hypothalamus and TNF-α, IL-1β and IL-6 in the serum were determined by ELISA. Finally, the plasma and urine samples of febrile rats were analyzed in combination with UHPLC-Q-TOF-MS to explore the antipyretic mechanism of IN. Results Compared with pyrexia model rats, the rectal temperature of rats treated with Indigo Naturalis was suppressed after 0.5 h (P <0.01). The enzyme-linked immunosorbent assay (ELISA) showed that the expression of tumor necrosis TNF-α (P <0.01) and IL-6 (P <0.05) were suppressed by IN. A total of 11 endogenous metabolites were identified from the plasma samples of the IN group, the model group and the control group. Furthermore,15 metabolites were identified in the urine samples. Pathway analysis showed that IN mainly regulated glycerophospholipid metabolism, glucuronide mutual conversion and tryptophan metabolism. Conclusion This study clarified the exact effect of IN on infectious fever. It mainly works by regulating pyrogenic cytokines and glycerophospholipid pathways. The present research provided a deeper understanding of the traditional heat-clearing effect of IN. It provided a reference for the further scientific research of IN and the selection of antipyretic drugs in clinical practice.

Background. Allergic rhinitis (AR) is a type I allergic disease induced by IgE. Traditional Chinese medicine Saposhnikovia divaricata (Turcz.) Schischk (SD) has anti-inflammatory and antiallergic effects. Materials and Methods. AR model mice were constructed with ovalbumin (OVA) sensitization to observe the improving effect of SD treatment on AR by counting the number of sneezing and rubbing the nose, hematoxylin-eosin, periodic acid-Schiff, and toluidine blue stains. In addition, the allergy and inflammatory cytokines levels and inflammatory cells were observed by ELISA and Wright’s-Giemsa stain. The protein levels of the TLR4/TRAF6/NF-κB and IL-6/ROR-γt/STAT3 pathways were measured by immunohistochemistry, quantitative real-time PCR, and western blot. The intestinal microflora abundance in mice was observed by 16S rDNA high-throughput sequencing. Results. SD treatment inhibited the sneezing and rubbing times of the nose, decreased the degree of a dense arrangement of cells and mucosal swelling and the number of goblet and mast cells of nasal lavage fluid, reduced the levels of IgE, histamine, Leukotriene B4, IL-4, IL-5, TNF-α, IL-6, and IL-17, the eosinophils, neutrophils, and lymphocytes number, the LR4, TRAF6, IL-6, ROR-γt, and STAT3 mRNA levels, respectively, while, it increased the IL-2, IL-10, IFN-γ, and TGF-β1 proteins. SD treatment inhibited the NF-κB, p-STAT3, TLR4, TRAF6, and p-IκBα/IκBα proteins. Besides, the effects of OVA and SD treatments were significantly correlated with the abundance of intestinal microflora. The abundances of Cytophagales, Burkholderia, Alteromonadales, Lactococcus, and Clostridiaceae were changed in SD treatment on AR mice. Conclusions. This study provides a possibility that the improvement effect of SD treatment on allergies and inflammation in AR mice may be related to the TLR4/TRAF6/NF-κB and IL-6/ROR-γt/STAT3 pathways and intestinal microflora modulation.
