Nordamnacanthal potentiates the cytotoxic effects of tamoxifen in human breast cancer cells

Abstract: Tamoxifen (TAM) is the mainline drug treatment for breast cancer, despite its side effects and the development of resistance. As an alternative approach, in the present study a novel combination therapy was established through combining TAM with nordamnacanthal (NDAM) in order to investigate the additive effect of these drugs in MCF-7 human breast cancer cells. A significant dose-dependent reduction in cell viability and an increase in apoptosis were observed in the MCF-7 cells cotreated with TAM and NDAM comp… Show more

“…Here, Artonin E was seen to significantly reduce the viability of MCF-7 cells while increasing its rate of apoptosis in a concentration-dependent manner. The percentage of cells undergoing apoptosis after Artonin E treatment was greater than MCF-7 cells undergoing apoptosis after treatment with Tamoxifen, a standard agent as previously reported by Subramani et al26…”

“…Anthraquinones have immensely contributed to the development of effective anti-cancer drugs [ 20 ]. NDAM has been reported to be cytotoxic to several cancer cell lines based on the MTT assay [ 14 16 ]. Herein, NDAM was shown to be cytotoxic to all three breast cancer cell lines, MCF-7, MDA-MB231 and 4T1 cells.…”

“…The bioactivities of nordamnacanthal have been reported but are very preliminary. These reports claim that nordamnacanthal possess anti-viral, anti-microbial and cytotoxic effects [ 14 16 ]. The toxicity as well as the effectiveness of nordamnacanthal as an anti-cancer agent in an in vivo setting has not been reported yet.…”

“…28 Oxidative stress can cause permanent modification of genetic material 29 which represents the first step involved in mutagenesis, carcinogenesis and various disorders such as Alzheimer, Hungtinton's disease, diabetes, and Parkinson. 30,31 In the previous report, nordamnacanthal was found to enhance cytotoxic effect of tamoxifen in treating human 32 Damnacanthal 6 enhanced the expression of p21 and caspase-7 subsequently increased apoptosis in human breast cancer MCF7 cell. 33 In the present study, only three other major compounds screened for cytotoxic activities using MCF7 and 4T1 cell lines ( Table 5).…”
