Normal human endometrium in cell culture

Abstract: Separation of human endometrium into its epithelial and stromal components has been achieved through collagenase digestion and has permitted a study of these two cell populations under specific experimental culture conditions. The stromal cell populations showed a progesterone response, were easily handled in culture, and displayed a limited in vitro life span typical of human diploid fibroblasts. In contrast, epithelium only survived in short-term primary culture and showed no clear hormone response. High-den… Show more

“…Cells were resuspended in a 0.8% ammonium chloride solution at 37°C for 5 min to lyse the red blood cells. After the cells were washed, the stromal cells were separated from the epithelial cells by using modifications of selective attachment [11] and selective trypsinization methods [12]. Briefly, the cells were suspended in DMEM containing 2% fetal bovine serum (FBS) (Hyclone, Logan, UT) and an antibiotic-antimycotic mixture (final concentrations: 25 U/ml penicillin, 25 pzg/ml streptomycin, and 2.5 g/ml amphotericin B).…”

“…Endometrial stromal cells were cultured as described elsewhere (6)(7)(8). Briefly, the endometrium was minced and then enzymatically digested with 0.1% collagenase type I and 0.05% DNAse (Worthington Biochemical, Lakewood, NJ).…”

“…Previous studies had demonstrated similar rates of endometrial cell binding to commercially available LP9 PMCs when compared with PMCs derived from parietal peritoneum and ovarian surface epithelium (23).…”
