Notch signaling in the hematopoietic system

Abstract: Because of its multiple effects in tissue homeostasis and cancer, Notch signaling is gaining increasing attention as a potential therapeutic target. Notch proteins belong to a family of highly conserved cell surface receptors. Ligand binding leads to proteolytic cleavage of Notch receptors by the gamma-secretase complex, followed by translocation of the active intracellular Notch domain into the nucleus and transcriptional activation. Multiple genetic and pharmacological methods are available to inhibit or act… Show more

“…[1][2][3][4] Notch signaling plays multiple roles in hematopoiesis and is essential for the establishment of definitive hematopoiesis through the generation of hematopoietic stem cells, 5 as well as for their subsequent differentiation in an expanding number of blood cell types. [6][7][8][9] The role of Notch signaling has been particularly well documented in T-cell development where Notch1/Dll4 interactions are crucial to induce T-lineage differentiation at the expense of other hematopoietic lineages. [10][11][12][13][14] Subsequently, Notch signaling is implemented in TCR-rearrangements, 15,16 modulation of TCR-αb versus -gd development, [17][18][19][20][21] and in the support of proliferation during b-selection.…”

“…Therefore, dysfunction of Notch prevents differentiation, ultimately guiding undifferentiated cells toward malignant transformation. Investigations of two last decades show that those disorders of the Notch signaling pathway are associated with development of some human haematological and solid cancers (1)(2)(3)(4)(5).…”

“…Interaction of Notch receptors with their ligands initiates the release of the active intracellular domain of Notch by proteolytic cleavages from the membrane, allowing its translocation into the nucleus. Once there, the intracellular domain of Notch forms a complex with recombination signal-binding protein-J, converting it from a repressor to an activator of transcription (8). There is increasing evidence revealing an important role for Notch signaling in the regulation of CD4 + Th cell differentiation or function in the periphery (9)(10)(11).…”
