NOTES: The DAL81 Gene Product Is Required for Induced Expression of Two Differently Regulated Nitrogen Catabolic Genes in Saccharomyces cerevisiae

Abstract: We demonstrate that the DAL81 gene, previously thought to be specifically required for induced expression of the allantoin pathway genes in Saccharomyces cerevisiae, functions in a more global manner. The data presented show it to be required for utilization of 4-aminobutyrate as a nitrogen source and for 4-aminobutyrate-induced increases in the steady-state levels of UGA1 mRNA. The DAL81 gene encodes a 970-amino-acid protein containing sequences homologous to the Zn(II)2Cys6 motif and two stretches of polyglu… Show more

“…Tup1 and Ssn6 form a corepressor complex in yeast and have a known role in regulating amino acid permeases (Tanaka and Mukai 2015), while Stp2 is an amino acid regulated TR that regulates amino acid permeases (Martínez and Ljungdahl 2005). Dal81 has a known role in positively regulating nitrogen degradation in S. cerevisiae (Bricmont et al 1991), but has a different role in regulating arginine synthesis in C. parapsilosis (Turner et al 2018).…”

“…Although the DBD’s metal biding portion is essential for the DNA binding activity, it can be dispensable in some cases and still exerts its regulatory activity [ 15 ]. In particular, the expression of a truncated-zinc Zn 2 Cys 6 finger motif DAL81 allele in a dal81 S. cerevisiae strain recovers its ability to induce the urea amidolyase activity, encoded by the Dal81p-regulated DUR1 , 2 gene [ 47 ]. The same was observed when the expression of a truncated DAL81 allele lacking the six cysteine residues of the Zn 2 Cys 6 finger motif could recover the ability of a dal81 mutant to express the β-galactosidase reporter under the control of the GABA upstream activation sequence (UAS GABA ), which is present in the promoters of both Dal81p- and Uga3p-regulated genes UGA1 and UGA4 [ 48 ].…”

“…Induction of this permease requires at least two positive acting proteins, the specifc Uga3p factor and the pleiotropic Uga35p factor (i.e. Dal81p/DurLp) [14,15]. These factors act through a 19 bp CG-rich upstream activating sequence, UAS GABA .…”
