Novel AKT1-GLI3-VMP1 Pathway Mediates KRAS Oncogene-induced Autophagy in Cancer Cells

Abstract: Background: Autophagy plays a role in cancer development. Results: Oncogenic KRAS induces Vacuole Membrane Protein 1 (VMP1) through a novel AKT1-GLI3-p300 pathway and requires VMP1 to regulate autophagy in cancer cells. Conclusion: Define a novel pathway initiated by the oncogene KRAS regulating autophagy. Significance: These findings contribute to the understanding of the mechanism underlying oncogene-induced autophagy.

“…In addition, about 13 GO downstream effectors such as mammalian target of rapamycin, KRAS indirectly modulates cell survival. 25 Through the Braf/ mitogen-activated protein kinase pathway, it also influences cell proliferation. 26 As part of a negative-feedback regulation, SOCS1 downregulates cytokine signaling through direct inhibition of the JAK tyrosine kinase and the signaling cascade of activated cytokine receptors, thereby attenuating cytokineinitiated signal transduction.…”

“…It is interesting to note that the expression of VMP1 is also transcriptionally activated by the mutated KRAS oncogene through a mechanism dependent on GLI3 and p300 (24). The KRAS oncogene possibly induces VMP1 expression to meet the increased energy needs of the cell during the transformation process.…”
