Novel antiangiogenic agents in dermatology

Abstract: Because angiogenesis underlies the pathogenesis of numerous conditions (cancer, psoriasis, macular degeneration), there is a pressing need for continued investigations into angiogenic signaling and potential drug targets. Antiangiogenic agents can be classified as either direct or indirect. Direct antiangiogenics act on untransformed endothelial cells to prevent differentiation and proliferation; indirect antiangiogenics act to inhibit factors involved in proangiogenic signaling. Agents currently available wit… Show more

“…Although the cellular mechanisms were not characterized, it appears that the main SPF treatment effects we observed could be attributed to reduced wound retraction in the first 12 days followed by inhibition of exuberant granulation tissue, especially within the first 30 days of wound healing. It is known that excessive inflammation can impair wound healing, and ionic silver, gentian violet, and methylene blue have been proposed to have anti‐inflammatory effects . Gentian violet and methylene blue have antiangiogenic properties, which could conceivably impede exuberant granulation tissue formation.…”

“…Because secreted VEGF is a major angiogenic factor in glioma, VPA inhibition of VEGF secretion in vitro and VEGF expression in vivo in the VPA Inhibits Glioma Angiogenesis brain are important actions of indirect angiosuppression of VPA. 1,4) The direct mechanism represents a direct effect on endothelial cells in terms of endothelial cell proliferation and tube formation. Second, combination of other anti-angiogenic agents, such as metronomic CPT-11 treatment, enhanced the anti-angiogenic and anti-tumor effects of VPA.…”

“…Studies show these compounds to be potent and efficacious inhibitors of Nox2 and Nox4 activity [61]. A structure-based approach to develop more specificity led to the synthesis of the fulvene derivatives class of inhibitors [62]. Fulvenes are aromatic molecules that have the advantage of being highly water soluble.…”
