Novel Antimicrobial Peptides from the Arctic Polychaeta Nicomache minor Provide New Molecular Insight into Biological Role of the BRICHOS Domain

Abstract: Endogenous antimicrobial peptides (AMPs) are among the earliest molecular factors in the evolution of animal innate immunity. In this study, novel AMPs named nicomicins were identified in the small marine polychaeta Nicomache minor in the Maldanidae family. Full-length mRNA sequences encoded 239-residue prepropeptides consisting of a putative signal sequence region, the BRICHOS domain within an acidic proregion, and 33-residue mature cationic peptides. Nicomicin-1 was expressed in the bacterial system, and its… Show more

“…Nicomicins consist of 33 residues (Table 1), containing BRICHOS domain in the sequences of their prepropeptide. They are characterized by many hydrophobic amino acids (51%) and a disulfide bond (Cys24 – Cys29) [110]. While Nicomicin-2 has no effect on bacteria, Nicomicin-1 exerts strong antimicrobial activity towards Gram-positive bacteria by damaging their membranes; the presence of salt impedes its activity [110].…”

“…Polychaete is a marine worm that belongs to phylum Annelida. It is a small invertebrate that inhabits all places in a marine environment and contributes to a high percentage of the total macrobenthic community diversity, abundance, and biomass [ 8 , 9 ]. In Malaysia, 64 species of polychaete that come from 31 families have been recorded from 1866 to 2013 [ 10 ].…”

“…Papers included in this Special Issue deal with marine polychaeta, providing good examples of their biological potential. The peptides arenicins and nicomicins, isolated from Arenicola marina [21,22] and Nicomache minor [23], respectively, exhibited in vitro antimicrobial activity and possessed cytotoxicity against cancer cells. Arenicin was shown to modulate the human complement system [22].…”
