Novel Blood Vascular Endothelial Subtype-Specific Markers in Human Skin Unearthed by Single-Cell Transcriptomic Profiling

Abstract: Ample evidence pinpoints the phenotypic diversity of blood vessels (BVs) and site-specific functions of their lining endothelial cells (ECs). We harnessed single-cell RNA sequencing (scRNA-seq) to dissect the molecular heterogeneity of blood vascular endothelial cells (BECs) in healthy adult human skin and identified six different subpopulations, signifying arterioles, post-arterial capillaries, pre-venular capillaries, post-capillary venules, venules and collecting venules. Individual BEC subtypes exhibited d… Show more

“…Nevertheless, besides the EPC cluster C7, we characterized 7 distinct EC signatures expressing gradually overlapping markers for arterioles (C1, C2), capillaries (C3, C4, C9), and postcapillary venules (C5, C6). This is in line with scRNAseq data from dermal ECs identifying 5 clusters (arterioles, capillary 1, capillary 2, postcapillary venules, venules) 71 , and 6 clusters (arterioles, post-arterial capillaries, prevenular capillaries, postcapillary venules, venules, collecting venules) 72 .…”

“…It is also known that miR-709 silences EGR2 by binding to the 3′UTR and the promoter region of EGR2 32 . Meanwhile, EGR2 is expressed in venular endothelial cells, but not in the endothelial cells of arterioles and post-arterial capillaries, based on an analysis of single-cell transcriptomic profiling for human dermal blood endothelial cells 33 . These suggest that miR-709 is expressed in arterial, but not in venous, endothelial cells.…”

“…For endothelial cell modules, gene sets defining tip, stalk, arteriole, venule, lymphatic, capillary and hypoxia scores (Extended data Fig. 9g, h) were derived from published literature [125][126][127] . The list of genes for each gene module is provided in Supplementary Tables 18, 20.…”
