Novel c-CBL and CBL-b ubiquitin ligase mutations in human acute myeloid leukemia

Abstract: The CBL ubiquitin ligase targets a variety of activated tyrosine kinases (TKs) for degradation. Many TKs are mutationally or autocrine activated and/or often overexpressed at the mRNA and protein levels in acute leukemias. We hypothesized that CBL is mutated in patients with acute myeloid leukemia (AML). IntroductionThe Casitas B-cell lymphoma (CBL) gene gives rise to a protein with multiadaptor function and E3 ubiquitin ligase activity that targets a variety of tyrosine kinases (TKs) for degradation. 1-3 c-C… Show more

“…Recent reports have shown that mutations in the Cbl family RING finger domain are not only associated with preleukemic chronic myelomonocytic leukemia, juvenile myelomonocytic leukemia, and other myeloproliferative neoplasms, but also with progression to AML (31,32). It is well known that the inhibition of PI3K/Akt activation by Cbl-b is dependent on a functional Cbl RING finger (18)(19)(20)(21)(22).…”

“…CBL mutations have been found in various myeloid malignancies, such as MDS, AML, and chronic myelomonocytic leukemia (CMML) [33][34][35]. CBL mutations have also been identified in the patients with FPD/AML.…”
