Novel cis-acting elements in petunia Cab gene promoters

Abstract: In order to identify specific cis-acting elements which regulate the expression of the divergent Cab22R and Cab22L genes of Petunia, we conducted systematic mutational studies of the 1 kb intergenic promoter region. Sequence analysis revealed three GATA box sequence repeats positioned between the TATA and CAAT box elements. These GATA elements are conserved in corresponding promoter regions of all LHCII Type I Cab genes in Petunia and other dicotyledonous plants we have examined. Site-specific mutations in the… Show more

“…Since a cab gene (P. thunberghii Cab 6) containing only modules 1 and 4 seems to be nonphotoregulated (Kojima et al, 1992(Kojima et al, , 1994, the minimal D N A modular array, which is consistently associated with light-responsive regions, is the one constituted by modules 1 , 2, and 4. Individual conserved DNA modules are: 1 to 3, GATA motifs (Gidoni et al, 1989); 4, CCAAT box; 5 , motif related to the ABF-2-binding site (Argüello et al, 1992) and contained within the region bound by CA-1, CUF2, and CUF3 factors (Sun et al, 1993;Carré and Kay, 1995); 6, CUF-1 binding-site (Anderson et al, 1994).…”

“…The latter display distinct "broadened" consensus in dicotyledons, which are RGATGAGATAAGAT (20 genes from species belonging to 10 genera and five families compared; not shown) and TGGTGNNYAAYGATAAGG (derived from 11 genes from plants belonging to four dicotyledonous families) for rbcS-CMA5 and rbcS-CMA4 IB modules, respectively. Cab-CMA1 also includes an IB element, which is part of a conserved array of three GATA motifs found in many cab gene promoters (Gidoni et al, 1989) and in which the consensus in angiosperms is GA-TANNGATAN(6-8)GATAAGR (Fig. 2).…”

“…The second motif is the light-responsive element GATA (-384 to -387). This sequence regulates transcription of both rhcS (small subunit of ribulose-1,5-bisphosphate carboxylase) and Cub (chlorophyll a/b binding protein) and is present in the promoter region of many genes essential for photosynthesis [23]. There are nine introns in the AKT3 genomic sequence, ranging in size from 79 to 256 bp (Table I).…”

“…The TATA element beginning at nt 524 deviates from the consensus TATA box defined by Bucher (1990), but is similar to an artificial TATA box construct that results in low RNA polymerase II activity in vitro in the absence of other cis -acting elements (Wang et al 1996) and is also similar to the TATA box associated with the petunia cab22R gene (Gidoni et al 1989). The low activity of these TATA boxes suggests a plausible reason why only a small number of transcripts initiate downstream of TATA 524 at nt 561.…”
