Novel CRISPR-based detection of Leishmania species

Abstract: Tegumentary leishmaniasis, a disease caused by protozoan parasites of the genus Leishmania, is a major public health problem in many regions of Latin America. Its diagnosis is difficult given other conditions resembling leishmaniasis lesions and co-occurring in the same endemic areas. A combination of parasitological and molecular methods lead to accurate diagnosis, with the latter being traditionally performed in centralized reference and research laboratories as they require specialized infrastructure and op… Show more

“…These methodologies present many advantages since they increase sensitivity by the fluorescent read-out and specificity conferred by the guide crDNA or crRNA. For example, CRISPR-Cas 13 (SHERLOCK) [ 109 ] or CRISPR-Cas12 (HOLMES: a one-Hour Low-cost Multipurpose highly Efficient System) [ 110 ] were tested with success on CL and HAT clinical samples [ 111 , 112 ]. These methodologies detect Nucleic acid but require an initial step for amplifying the genetic material (RNA or DNA) by isothermal or classical PCR methodologies.…”

“…Additionally, the CRISPR-Cas12a system has applications in the molecular identification of Leishmania spp. Common multi-copy target assays, such as PCR for leishmaniasis molecular diagnostics, include the highly conserved 18S ribosomal RNA gene (18S rDNA) and a segment of kinetoplast DNA (kDNA) minicircles ( Dueñas et al, 2022 ). Despite the establishment of various diagnostic techniques, the scope for detection of parasite by developing more sensitive techniques is crucial.…”
