Novel Dermal Delivery Cargos of Clobetasol Propionate: An Update

Abstract: Dermal disorders such as psoriasis and eczema are associated with modifications in the chemical and molecular composition of the skin. Clobetasol propionate (CP), a superpotent topical glucocorticoid, is widely used for the therapeutic management of various skin conditions, owing to its strong anti-inflammatory, antipruritic, vasoconstrictive, and antiproliferative activities. Safety studies demonstrated that CP is safer for a shorter period, however, with prolonged application, it shows secondary side effects… Show more

“…CLO, a prednisone predecessor, is the most frequently recommended steroidal antipsoriatic medication. 92 , 93 A recent study has indicated notable improvements in the unique characteristics of psoriasis-associated lesions with CLO treatment. These positive outcomes may potentially be attributed to the medication's anti-inflammatory, immunosuppressive, apoptotic, vasoconstricting, and anti-proliferative activities.…”

“…Microemulsions (MEs) are homogenous in appearance, single-phase, clear, and thermodynamically steady colloidal mixtures consisting of water, oil, co-surfactant, and surfactant, with droplets typically ranging up to 100 nm in size. Though a microemulsion has a smaller droplet size compared to Nes, both terms are still being used [ 54 ]. MEs present a promising platform for drug delivery, offering improved drug solubility, stability, absorption, and targeted delivery for enhanced therapeutic outcomes.…”

“…CP is a high-potency synthetic GC widely used in dermatology to treat different dermatoses including psoriasis, 1 eczema, 2 lichen planus, 3 and seborrheic dermatitis. 4 Antiproliferative, immunosuppressive, vasoconstrictive, and anti-inflammatory actions of CP account for its broad therapeutic applicability; 5 however, at the same time, these features cause multiple local and even systemic adverse effects with long-term use. 6,7 Moreover, lipophilicity and poor aqueous solubility of CP result in its low permeation through the skin affecting its dermal bioavailability.…”

“…To date, a wide range of drug delivery systems have been proposed for topical CP administration, including the conventional forms, such as creams, ointments, and foams, 9 as well as more advanced ones, like polymer- and lipid-based vehicles. 5 Among the conventional CP formulations currently available on the market, Clobevate, Embeline, and Temovate are some of the most broadly used ones. However, it is also worth noting that the use of available topical forms can be inconvenient.…”
