Novel INHAT Repressor (NIR) is required for early lymphocyte development

Abstract: Novel inhibitor of histone acetyltransferase repressor (NIR) is a transcriptional corepressor with inhibitor of histone acetyltransferase activity and is a potent suppressor of p53. Although NIR deficiency in mice leads to early embryonic lethality, lymphoidrestricted deletion resulted in the absence of double-positive CD4 + CD8+ thymocytes, whereas bone-marrow-derived B cells were arrested at the B220 + CD19− pro-B-cell stage. V(D)J recombination was preserved in NIR-deficient DN3 double-negative thymocytes, … Show more

“…Notably, the autoimmune disease-causal variants were more enriched in the binding sites with blood cell-specific cooperation evidence (3DCoop-specific) than the binding sites with unrestricted cooperation evidence (3DCoop-unrestricted) for each of the 102 selected TRs ( p = 2.41e-07, one-tailed paired t test, Figure 5 D). For example, GFI1, IKZF5, and NOC2L are highly expressed factors in GM12878 cells and are essential TRs in lymphopoiesis ( Ma et al., 2014 ; Merkenschlager, 2010 ; van der Meer et al., 2010 ). We found that, compared with the randomly sampled control variants, the autoimmune disease-causal variants exhibited similar binding enrichment patterns among these TRs, where the binding sites with blood cell-specific cooperation evidence received the best enrichment rather than those inferred from other strategies ( Figure 5 E).…”

“…Based on the knowledge of the biological properties and function of NOC2L, further investigation was conducted to examine how NOC2L affects biological events. It has been proposed that NOC2L can participate in some key biological events, including the rRNA biogenesis [ 48 ], embryonic development [ 49 , 50 ], and cancer progression [ 9 , 51 ]. All these biological events affected by NOC2L can be contributed to its characteristic and function mentioned previously in this review.…”

“… 6 NIR has been found to play important functions in lymphocytes development by cooperating with p53 to impose checkpoint for the generation of mature B and T lymphocytes. 7 But the role of NIR in breast cancer is unclear.…”
