NPAS1 Represses the Generation of Specific Subtypes of Cortical Interneurons

Abstract: Summary Little is known about genetic mechanisms that regulate the ratio of cortical excitatory and inhibitory neurons. We show that NPAS1 and NPAS3 transcription factors (TF) are expressed in progenitor domains of the mouse basal ganglia (subpallium, MGE and CGE). NPAS1−/− mutants had increased proliferation, ERK signaling and expression of Arx in the MGE and CGE. NPAS1−/− mutants also had increased neocortical inhibition (sIPSC and mIPSC), and generated an excess of somatostatin+ (SST) (MGE-derived) and vaso… Show more

“…At E17.5, Npas1 expression was reduced, a TF that marks SST + but not PV + interneurons (Fig. S3H,I) (Stanco et al, 2014). Therefore, at E12.5 Coup-TF2-sensitized mutants generated fewer immature interneurons expressing Sst, Lhx6 and Sox6.…”

“…By 17.5, mutants showed reduced expression of Npas1 (Fig. S3H,I), a TF that marks SST + and not PV + interneurons, and that positively regulates SST + interneuron numbers (Stanco et al, 2014). The dMGE has been proposed to be a major source of SST + interneurons (Flames et al, 2007;Wonders et al, 2008;Inan et al, 2012 (A,B) and Coup-TF2-sensitized mutant (A′,B′) coronal hemisections in two rostral-caudal planes (n=3 for each control and mutant).…”
