Nrf2-Inducing Anti-Oxidation Stress Response in the Rat Liver - New Beneficial Effect of Lansoprazole

Abstract: Lansoprazole is a potent anti-gastric ulcer drug that inhibits gastric proton pump activity. We identified a novel function for lansoprazole, as an inducer of anti-oxidative stress responses in the liver. Gastric administration of lansoprazole (10–100 mg/kg) to male Wistar rats produced a dose-dependent increase in hepatic mRNA levels of nuclear factor, erythroid-derived 2, -like 2 (Nrf2), a redox-sensitive transcription factor, at 3 h and Nrf2 immunoreactivity (IR) in whole hepatic lysates at 6 h. Conversely,… Show more

“…Nrf2, a redox sensitive transcription factor, is a fundamental contributor to oxidative stress homeostasis [56]. Nrf2 is involved in the regulation of the synthesis and conjugation of glutathione (glutamate-cysteine ligase catalytic subunit), and antioxidant proteins responsible for the detoxification of ROS [57]. It was reported previously that Nrf2 expression is significantly lower in semen of men with low sperm motility [58].…”

“…To strongly confirm whether Nrf2 is responsible for SULT1E1 expression we also did an in vitro experiment with lansoprazole, a known Nrf2 inducer. Lansoprazole a potent gastric ulcer drug which inhibits proton pump induces anti-oxidative stress via induction of Nrf2 [73]. The cells incubated with lansoprazole showed an elevated SULT1E1.…”

“…HO‐1 is also called heat shock protein 32, which can provide cytoprotection against oxidative stress . Induction of HO‐1 is controlled by Nrf2 . Modulation of the Nrf2 system is a potential pharmacological target for ameliorating oxidative stress.…”
