Nrf2-Mediated Dichotomy in the Vascular System: Mechanistic and Therapeutic Perspective

Abstract: Nuclear factor-erythroid 2-related factor 2 (Nrf2), a transcription factor, controls the expression of more than 1000 genes that can be clustered into different categories with distinct functions ranging from redox balance and metabolism to protein quality control in the cell. The biological consequence of Nrf2 activation can be either protective or detrimental in a context-dependent manner. In the cardiovascular system, most studies have focused on the protective properties of Nrf2, mainly as a key transcript… Show more

“…Most cancers show elevated levels of ROS and oxidative stress, which can cause DNA damage, impair protein function, and alter mechanisms of cellular proliferation to promote tumorigenesis [ 154 ]. This is particularly true for cancers of the skin and lung, as both organs are directly exposed to additional sources of environmental ROS which contribute to cancer initiation and aggravation [ 158 , 159 ].…”

“…Furthermore, not all actions of Nrf2 are good for the body. Nrf2 may promote tumours [ 135 , 136 ] and atherogenesis [ 137 , 138 ], so any drug acting on Nrf2 must avoid these two and maybe other unknown potentially harmful actions.…”

“…Aging and Disease • Volume 15, Number 5, October 2024 NQO1, heme oxygenase-1 (HO-1) and ferritin, to maintain redox homeostasis [68]. Stimulation of Nrf2 can have positive effects on the endothelium and powerful anti-inflammatory and pro-angiogenic effects on the developing vascular system [69][70][71]. Compelling evidence supports the theory that Nrf2 dysfunction in the aging vascular system worsens oxidative stress and reduces the ability of vascular cells to withstand ROSmediated cytotoxicity.…”
