Nucleus accumbens AMPA receptor trafficking upregulated by food restriction: an unintended target for drugs of abuse and forbidden foods

Kenneth D Carr

Abstract: There is a high rate of comorbidity between eating disorders and substance abuse, and specific evidence that weight-loss dieting can increase risk for binge pathology, rebound excessive weight gain, and initiation and relapse to drug abuse. The present overview discusses basic science findings indicating that chronic food restriction induces dopamine conservation, compensatory upregulation of D-1 dopamine receptor signaling, and synaptic incorporation of calcium-permeable glutamatergic AMPA receptors in nucleu… Show more

“…Thus, these results also suggest that the mesolimbic dopamine system is not responsible for increasing VS*, but approach motivation, a notion that is consistent with inferences derived from a review of other studies (Ikemoto and Panksepp 1999). The finding that CFR 3wk interacts with MPH in increasing approach motivation is consistent with previous studies suggesting that CFR sensitizes the mesolimbic dopamine system for motivational effects of psychostimulant drugs (Carr 2016; Pothos et al 1995; Stouffer et al 2015). …”

“…The brain levels of dopamine in males can be controlled by an intake of tyrosine in food, and chronic food restriction induces dopamine conservation (31). However, in the …”

“…While to our knowledge the current study is the first attempt to examine reinforcing value of non-nutritive orosensory stimulation in people, Epstein and Raynor have shown food deprivation to increase the relative reinforcing value of food in a computer choice paradigm similar to our own (Raynor & Epstein, 2003). Among laboratory animals, increased appetitive responding toward various drugs of abuse has repeatedly been demonstrated in chronically food-restricted animals (Cabeza de Vaca & Carr, 1998), as has been speculated to put food-restricted people at risk of binge eating episodes (Carr, 2016). In the animal model of sham feeding, reducing the duration of food deprivation limits the duration of intake, supporting a relationship at least between short-term food restriction and drive for sweet reward (Weingarten, Duong, & Elston, 1996).…”
