Numerous post-translational modifications of RNA polymerase II subunit Rpb4 link transcription to post-transcriptional mechanisms

Abstract: Rpb4/7 binds RNA Polymerase II (Pol II) transcripts co-transcriptionally and accompanies them throughout their lives. By virtue of its capacity to interact with key regulators (e.g., Pol II, eIF3, Pat1) both temporarily and spatially, Rpb4/7 regulates the major stages of the mRNA lifecycle. Here we show that Rpb4/7 can undergo over 100 combinations of post-translational modifications (PTMs). Remarkably, the Rpb4/7PTMs repertoire changes as the mRNA/Rpb4/7 complex progresses from one stage to the next. A mutage… Show more

We demonstrated that Pol II regulates mRNA translation and decay by mediating Rpb4/7 cotranscriptional binding to Pol II transcripts [10][11][12] , a process we named "mRNA imprinting" 13,14 .More "classical" yeast DFs -components of the Ccr4-Not complex -also imprint mRNA and regulate mRNA export, translation and decay 15,16 . Thus, cellular localization of these factors seems to be critical for their two opposing activities, either in synthesis or degradation. Here we show that Xrn1 nuclear import, mediated by two NLSs and by the decaying RNA, affects mRNA synthesis and decay of a large portion of the mRNAs.…”
