Nutlin-3 preferentially sensitises wild-type p53-expressing cancer cells to DR5-selective TRAIL over rhTRAIL

Abstract: Background: Tumour cell-selective activation of apoptosis by recombinant human TNF-related apoptosis-inducing ligand (rhTRAIL) is enhanced through co-activation of p53 by chemotherapeutic drugs. The novel anticancer agent nutlin-3 provides a promising alternative for p53 activation by disrupting the interaction between p53 and its negative feedback regulator MDM2.

“…Consequently, tissue slices became an attractive alternative to immortalized cell lines since at the time it was widely recognized that there is a need for more complex model systems. In the last decades, precision‐cut slices from various patient tumors such as carcinomas from breast [134–139], lung [138, 140–142], brain [143, 144], ovary [145, 146], cervix [147], kidney [148], and prostate [149] were introduced and used in various fields of cancer research.…”

“…Successful infections of cancer cells in tissue slices with different viruses have been described showing that this ex vivo model is also suitable for the validation of alternative anti‐cancer approaches such as oncolytic viral infection or gene therapy [143, 156–160]. In addition, tissue slice culture has been used for validation purposes in studies utilizing immortalized cell lines from different tumor entities grown in 2D [139, 145, 148]. Meanwhile, drug testing services based on patient‐derived tumor tissue slices are commercially available (http://indivumed.com).…”

“…Notably, 9 of the 18 genes were associated with neoplasms in their annotations. Examples include TNFRSF10B, inducing apoptosis in various solid tumors, [15][16][17][18][19] PTPN13 being upregulated in pancreatic cancer and Ewing sarcoma, 20,21 or IER3 and CCR4 that have a role in myelodysplastic syndromes and lymphoma, respectively. 22,23 In addition, TP63 contributes to tumorigenesis in several cancers.…”

“…We assume that this increase in p21 following treatment with doxorubicin is due to the increase in p53 we observed and occurs by the pathways that have previously been documented in colon cancer cell lines [1720]. Mdm2 inhibitors, such as Nutlin-3 and MI-43, increase the expression of p53, p21, Mdm2 and DR5 expression in p53 wild type colon cancer cell lines [2021]. The compounds, YH263, YH264 and WW751, did not increase the expression of Mdm2.…”
