Occurrence and Genetic Diversity of Uncultured Legionella spp. in Drinking Water Treated at Temperatures below 15°C

Abstract: Representatives of the genus Legionella were detected by use of a real-time PCR method in all water samples collected directly after treatment from 16 surface water (SW) supplies prior to postdisinfection and from 81 groundwater (GW) supplies. Legionella concentrations ranged from 1.1 ؋ 10 3 to 7.8 ؋ 10 5 cells liter ؊1 and were significantly higher in SW treated with multiple barriers at 4°C than in GW treated at 9 to 12°C with aeration and filtration but without chemical disinfection. No Legionellae (<50 CFU… Show more

“…Notwithstanding, L. pneumophila was detected in both streams (surface water) and wells (ground water). This study is comparable to a study conducted in Netherlands where PCR technique was used for detection of legionellae in groundwater (well) and surface water (stream); surface water showed a high yield of Legionella organism compared to groundwater water samples (Wullings and van der Kooij, 2006).…”

“…in water systems. A large diversity of Legionella spp., many of which are as yet uncultured, is ubiquitous in freshwater environments at temperatures of Ͻ20°C (36,42,43,44,45,46). L. pneumophila is not cultured or only sporadically cultured from these low-temperature environments.…”

“…Water temperatures in supplies A and B were below 20°C and thus were too low for growth of L. pneumophila (32), but uncultured Legionella species, including Legionella-like amoeba pathogens (82), can multiply in this temperature range in water supplies. A number of the detected free-living protozoa can probably serve as hosts for these uncultured Legionella bacteria.…”
