Staphylococcus aureus is a prevalent bacterial colonizer with zoonotic potential, affecting humans and various animal species, including livestock, poultry, and pets. This study investigates methicillin-resistant S. aureus (MRSA) isolates in poultry meat portions using Multiplex PCR methods. A total of 210 samples, including 70 wings, 70 thighs, and 70 necks, were collected from Mashhad, Iran markets. S. aureus identification employed culture and phenotypical methods, while the disk diffusion method assessed antibiotic susceptibility using 14 different disks. The Multiplex PCR assay was developed to confirm S. aureus isolates and detect antibiotic resistance genes. Among the 210 samples, 52 (24.76%) tested positive for S. aureus. Antibiotic susceptibility testing revealed that 17 (32.69%) of the S. aureus isolates were resistant to methicillin. Tetracycline exhibited the highest resistance, followed by ampicillin (61.5%) and penicillin (57%). Conversely, chloramphenicol demonstrated the lowest resistance at 3.8%. All isolates were susceptible to gentamicin, vancomycin, imipenem, and ciprofloxacin. PCR analysis confirmed the presence of 16S rRNA and femA genes in all isolates, while 14 (26.92%) harbored the methicillin-resistant gene (mecA). The study suggests multiplex PCR is a valuable and sensitive technique for detecting antibiotic resistance genes in S. aureus within chicken meat, emphasizing its utility in surveillance and control efforts.

The study investigated the economic concerns associated with livestock-associated methicillin-resistant Staphylococcus aureus (LA-MRSA) in livestock (cow), examining its connection to severe infections, antimicrobial resistance (AMR), and virulence factors. The research, conducted in Edo State, Nigeria, analyzed 400 samples (200 rectal and 200 nasal swabs) collected between March 2018 and February 2019. MRSA prevalence was identified using conventional culture-based methods and polymerase chain reaction (PCR) techniques, revealing 63.5% (n = 254) for Staphylococcus aureus and 55% (n = 220) for MRSA. Of the 76 mecA-positive MRSA isolates, 64.5% (n = 49) exhibited multidrug resistance (MDR) while the remaining were sensitive to specific antimicrobials. Key virulence genes, such as PVL (81.6%; n = 62) and tsst-1 (44.7%; n = 34), were prevalent, along with AMR genes like mecC, tetM, ermA, ermC, vanA, and vanC. Staphylococcal chromosomal cassette mec (SCCmec) typing identified different types, notably II, IVa, and IVb. Biofilm formation, a crucial virulence factor varied in strength, is associated with icaA and icaB genes (p < 0.01). The findings highlighted substantial AMR and biofilm-forming capacity within LA-MRSA isolates, emphasizing the importance of ongoing surveillance for informed treatment strategies, AMR policies, and control measures against MDR staphylococcal infections.

The irrational use of drugs, especially antimicrobials as growth promoters, and in the management of disease conditions without proper diagnosis has led to a higher incidence of multidrug resistance/antimicrobial resistance, which is a leading cause of death both in humans and animals. Improper diagnosis, irrational use of drugs and lack of adherence to withdrawal periods are factors incriminated in the incidence of multidrug resistance pathogens. This study was aimed at examining the prevalence, isolation, and antimicrobial susceptibility profile of Staphylococcus (S.) aureus in marketed milk and cheese in Ilorin, Kwara State, Nigeria. Using a cross-sectional study design which involved sampling of milk and cheese (n = 340) from four markets (Gambari, Ojaoba, Sango and Ipata) in Ilorin metropolis, Staphylococcus aureus (S. aureus) was isolated using standard microbiological procedures. Antimicrobial susceptibility testing of all positive isolates was done using the disk diffusion method. The prevalence of S. aureus was 3.5% in nono milk and 0.0% in cheese. The S. aureus isolated showed 100% resistance to ampicillin, ceftazidime and oxacillin and a high resistance rate to cefoxitin (91.7%), ceftriaxone (83.3%) and tetracycline (83.3%). Ciprofloxacin, gentamicin, and azithromycin showed lower resistance (25%) to the tested S. aureus isolates. The S. aureus isolates displayed six different resistance patterns and were resistant to three or more antibiotics. Particularly, three isolates were pan-resistant. This study emphasizes the importance of good management and hygiene practices throughout the milk processing value chain to ensure the quality and safety of the final product for consumers.
