Oct-4-expressing cells in human amniotic fluid: a new source for stem cell research?

Abstract: The results presented here suggest that human amniotic fluid may represent a new source for the isolation of human Oct-4-positive stem cells without raising the ethical concerns associated with human embryonic research.

“…Several authors have shown the presence of different SC subpopulations inside the amniotic fluid [1][2][3], but the isolation and characterization of CD117 pos (c-kit pos ) amniotic fluid stem (AFS) cells through immune selection is relatively recent [4,5]. We focused our attention on CD117 pos AFS cells that represent a subpopulation of fetal SCs expressing the type III-tyrosine kinase receptor of the stem cell factor (c-kit), and are considered as excellent candidates for the SC based-approaches in regenerative medicine.…”

“…Prusa et al (2004) observed the presence of a cell distinct population expressing POU5F1 (Oct4), a marker for pluripotent stem cells known to be expressed in ESCs, in human AF (Pesce & Scholer 2001). The mechanisms involved in the POU5F1 regulation in ESC have been widely investigated, but in the adult stage (including post-natal and fetus stages) POU5F1 function has not been identified even though it has been associated with the undifferentiated pluripotent state of stem cell populations derived from various adult human tissues or organs such as bone marrow-derived multipotent adult progenitor cells.…”

“…In addition, human amniotic epithelial (hAE) cells seem to be relatively resistant to rejection even after allotransplantation because of their expression of immunosuppressive factors, such as CD59 (Rooney and Morgan, 1992) and soluble HLA-G (Rebmann et al, 1999). Alternatively, it was suggested that amnion-derived cells may possess multipotency because human amniotic fluid cells contain a minor cell population expressing Oct-4 (Prusa et al, 2003). We also reported the expression of Oct-4 in cells derived from the human amnion (Wei et al, 2003).…”
