Oct4 upregulates osteopontin via Egr1 and is associated with poor outcome in human lung cancer

Abstract: Background Roles of cancer stem cells and early growth response gene 1 (Egr1) in carcinogenesis have been extensively studied in lung cancer. However, the role of Egr1 in the metastasis of lung cancer remains undetermined, especially in regard to stem cell-related pathways. Methods Egr1, osteopontin (OPN) and Oct4 expression in human lung cancer was determined by performing immunohistochemistry. Immunoblotting, ELISA, luciferase reporter assay, chromatin immunoprecipita… Show more

“…As such, it is emerging as an important cancer biomarker and its relevance in PI3K/Akt and integrin-NF-κB/HIF-1α pathways is currently a focus of ongoing research [ 93 , 94 , 95 , 96 , 97 , 98 ]. OPN expression in the context of cancer includes regulation by hyaluronic acid, IL-6, transmembrane protein TM4SF4, and Oct4-Egr1 pathway [ 94 , 99 , 100 , 101 ]. Angiogenesis, degradation of ECM, the formation of lamellipodia, and switching phenotypes towards either cancer-associated fibroblasts or stem cells are ways in which OPN can promote invasion and metastasis.…”

“…SPARC has been suggested as a prognostic biomarker for certain cancers such as soft tissue sarcoma, esophageal SCC and glioblastoma since its expression correlates with poor survival (232)(233)(234). In addition, SPP1 may be prognostic for breast, lung, gastric, liver and colon cancers since it associated with tumor progression and decreased patient survival (235)(236)(237)(238).…”

“…Lindahl's [36] study has shown that OPN can serve as an in ammatory mediator to the pathogenesis of breast cancer. Feng's [37] study shows that the upregulation of OPN is not conducive to the survival in human lung cancers. However, our study is the rst meta-analysis to discuss the relationship between survival and OPN expression, and we also evaluate the expression of OPN in different prostate tissues and different clinical parameters.…”
