Ocular Bioanalysis of Moxifloxacin and Ketorolac Tromethamine in Rabbit Lacrimal Matrix Using Liquid Chromatography–Tandem Mass Spectrometry

Amol Chhatrapati Bisen,
Rabi Sankar Bhatta

Abstract: Aim: The fixed-dose combination of moxifloxacin (MOXI) and ketorolac tromethamine (KTR) is widely used for the treatment of bacterial keratitis. Thus, a new LC–MS/MS method was developed to determine MOXI and KTR in lacrimal fluid. Methods: Bioanalysis was performed using a Shimadzu 8050 LC–MS/MS in electrospray ionization-positive mode and the method was validated per US FDA guidelines. Isocratic separation was performed with a Waters Symmetry C18 column using methanol and 0.1% formic acid containing deionize… Show more

“…Ulcerative keratitis is the most common and distinctive type of corneal blindness. [94][95][96][97] It is a spontaneous and long-lasting result of a microbial infection of the cornea, which is usually marked by ocular surface dystrophy. [98,99] Following post-operative surgery, trauma, and various ophthalmic procedures, the occurrence of bacterial and fungal co-infections can lead to the development of ulcers on the cornea.…”

“…These agents can be used standalone or in conjunction with anti-infective agents such as antifungal medication to reduce the pain incurred. For example: ketorolac tromethamine, diclofenac sodium, nepafenac, bromfenac etc. , Immunomodulators such as cyclosporin and lifitegrast are typically indicated following postoperative surgeries . However, their prolonged use may increase the risk of fungal comorbidities, which can potentially be life-threatening.…”
