On-demand nanozyme signal enhancement at the push of a button for the improved detection of SARS-CoV-2 nucleocapsid protein in serum

Abstract: We developed an innovative 3D printed casing that incorporates a lateral-flow immunoassay, dehydrated signal enhancement reagents, and a sealed buffer chamber.

“…In this device, a single button push is sufficient to enhance the signal after the LF immunoassay detection step. This device could detect SARS-CoV-2 infection at the early stages of infection (Bradbury et al, 2021).…”

“…Bradbury et al developed an all-in-one nanozyme signal-enhanced LFIA by using gold nanozyme with platinum coating as the signal label, which allowed the detection of N protein in serum at concentrations as low as 0.1 ng mL −1 . 59 The nanozyme probes can catalyze the oxidation of TMB reagent, finally leading to the formation of a purple product which can improve the sensitivity of the LFIA. Despite the successful development of a nanozymes-based LFIA assay for SARS-CoV-2, there is a need for further investigation into the underlying principle of enhanced nanozyme activity.…”

“…The selection of the capture probe is important for the construction of the sensor. Some specific recognition targets based on NP detection use molecular imprinting [32] , [33] , antigen-antibody reactions [34] , [35] , [36] , [37] , [38] , and aptamers [15] . Aptamers display more affinity and more binding sites with lower molecular weight and are easy to synthesize/modify, and cost-effective.…”
