On‐line 2D‐LC‐ESI/MS/MS determination of rifaximin in rat serum

Abstract: A highly sensitive and selective on-line two-dimensional reversed-phase liquid chromatography/electrospray ionization-tandem mass spectrometry (2D-LC-ESI/MS/MS) method was developed and validated to determine rifaximin in rat serum by direct injection. The 2D-LC-ESI/MS/MS system consisted of a restricted access media column for trapping proteins as the first dimension and a Waters C(18 )column as second dimension using 0.1% aqueous acetic acid:acetonitrile as mobile phase in a gradient elution mode. Rifampacin… Show more

“…At present, on-line 2D-LC systems, both LC-LC and LC × LC approaches, are considered powerful analytical tools, and many related applications have been reported in different fields [55,56]. CHMs, the mixture of numerous unknown components, were analyzed using these on-line systems with encouraging results, as seen from the data summarized in Table 2.…”

“…The literature shows methods for detection of rifaximin in biological fluid as rat urine, mouse serum, human plasma and dried blood spots and milk by HPLC or high performance liquid chromatography coupled to mass spectrometry (HPLC-MS). [10,[26][27][28][29][30][31][32] The literature also shows the detection of rifaximin in pharmaceutical product as tablets by spectrophotometry in the ultraviolet (UV), [33][34][35][36] visible (Vis) [37] and infrared region (IR), [38] capillary electrophoresis (CE), [39] thin layer chromatography (TLC) [40] high-performance thin-layer chromatographic (HPTLC) [41] ; HPLC [42][43][44][45][46][47][48][49][50] and HPLC-MS. [40] The literature also reports microbiological method for the calculation of the power of rifaximin by turbidimetry, [44] which is a more dynamic method than the traditional method by diffusion in agar. In the case of antimicrobials, the simultaneous evaluation of the material by physico-chemical and microbiological methods are extremely important.…”

“…However these methods have several limitations such as lack of selectivity, low sensitivity or large volume of biological matrix required. In our previous report (Rao et al , ), a restricted access media column (RAM‐Hisep) was used for the direct injection of biological fluids. In continuation of our work, it has become necessary to investigate a rapid microanalytical technique for therapeutic monitoring of of rifaximin in blood serum.…”
