Onconeural antigens and the paraneoplastic neurologic disorders: at the intersection of cancer, immunity, and the brain.

Abstract: Paraneoplastic neurologic disorders (PNDs) are believed to be autoimmune neuronal degenerations that develop in some patients with systemic cancer. A series of genes encoding previously undiscovered neuronal proteins have been cloned using antiserum from PND patients. Identification of these onconeural antigens suggests a reclassification of the disorders into four groups: those in which neuromuscular junction proteins, nerve terminal/vesicleassociated proteins, neuronal RNA binding proteins, or neuronal signa… Show more

“…The fixed tissue was placed in phosphate buffered saline (PBS) containing 2% Triton X-100 for roughly 6 hr and then placed in antibody diluent (PBS with 0.3% Triton X-100, 5.0% goat serum and 0.05% sodium azide) containing HNK-1 (ascites produced from cells obtained from American Type Culture Collection ([ATCC], Manassas, VA; 1/1,000), human anti-Hu (1/1,000; Fairman et al, 1995), and mouse anti-␤ (III)-tubulin (1/500; Lee et al, 1990a,b) for 12-24 hr at 4°C. Hu recognizes a family of RNA-binding proteins (Darnell, 1996;Okano and Darnell, 1997), and ␤ (III)-tubulin is an isoform of tubulin unique to neurons (Lee et al, 1990a,b). Tissues were washed and incubated in a mixture of goat anti-mouse immunoglobulin (Ig) M-Texas Red (1/100; -specific, Jackson Labs, West Grove, PA), goat anti-mouse CY2 (1/100; ␥ specific, Jackson Labs), and goat anti-human IgG-Alexa 488 (1/100; Molecular Probes, Eugene, OR).…”

“…Several autoantibodies associated with PND are directed against various nervous system antigens, including a group of proteins with potential roles in the signal transduction pathway (Darnell, 1996). The discovery of anti-CV2 autoantibodies allowed us to identify Ulip/CRMP proteins as potential targets for these autoantibodies (Honnorat et al, 1999).…”

“…High‐titer antibodies to the neuronal protein are found in both the serum and cerebrospinal fluid (CSF) of patients with PND 1. However, antibodies are unlikely to be the sole cause of the neuronal destruction for those PNDs in which the target antigens are intracellular 2, 3. Moreover, PND antigen‐specific CD8 + T cells are present in the peripheral blood and CSF of patients with both paraneoplastic cerebellar degeneration (PCD) and the paraneoplastic Hu syndrome subtypes of PND 4, 5, 6…”
