Ontogeny of Insulin-Like Growth Factors (IGF-I and IGF-II) and IGF-Binding Proteins in Porcine Serum during Fetal and Postnatal Development*

Abstract: The insulin-like growth factors (IGF-I and IGF-II) circulate in plasma in association with IGF-binding proteins (IGFBPs). As a first step to understanding the regulation of expression of these proteins in pigs, we characterized the ontogeny of circulating IGFs and IGFBPs during fetal and early postnatal development. Serum IGFs were separated from IGFBPs, before IGF RIA, by acidification and chromatography on C18 Sep-Pak cartridges. The IGF-I levels increased during the latter half of fetal life from 11 +/- 1 n… Show more

“…To examine how this protein might interact with and affect uterine cell function, a construct consisting of pIGFBP-2 cDNA in pET-15b expression vector was generated and used to produce recombinant protein in E. coli (Figs 1, 2). Consistent with the size of porcine IGFBP-2 (Lee et al 1991), the measured size of rpIGFBP-2 fused to the amino-terminal extension containing a hexa-histidine motif was 36 kDa (Fig. 2).…”

“…3) or Western blot analyses (data not shown). The highest-molecular-weight protein in GE CM appeared as a diffuse band between 53 and 45 kDa, which is consistent with the size of IGFBP-3 (Lee et al 1991, Ko et al 1994.…”

“…These data supported the absence of any obvious GH secretion deficiency in MS pigs. In accordance with previous works in pigs (Lee et al, 1991;Lamberson et al, 1995), the concentration of plasma IGF-I increased with age in LW pigs. At 80 days of age, plasma IGF-I concentrations did not differ between the two breeds in accordance with our previous observations taken between 10 and 150 days of age (Louveau et al, 1991).…”

“…Inter-and intra assay coefficients of variation were 6.8 and 6.25% for insulin and 6.0 and 4.43% for C-peptide, respectively. Plasma total IGF-I was measured by radioimmunoassay after acidification (1 glI00 mL trifluoroacetic acid) and chromatography (Sep-Pack Plus CIS) to remove the binding proteins (18). IGF-I antiserum was obtained courtesy of Drs.…”
