Optical Measurement of Three-Dimensional Collagen Gel Constructs by Elastic Scattering Spectroscopy

Abstract: Analysis of the formation and organization of new connective tissue formed in tissue-engineered constructs is a major requirement for tissue bioreactor technology. We have analyzed early-stage responses in collagen lattices, using elastic scattering spectroscopy to assess its potential to monitor tissue structural changes in structures up to 3 mm thick, under normal culture conditions. The method is based on an optical system in which an optical fiber delivers white light onto the tissue and the back-scattered… Show more

“…The optical setup used was that described in detail and schematically by Marenzana et al 13 In brief, it consisted of a xenon white light source, a fiber optic probe, and a spectrometer (Ocean Optics, Dunedin, FL) and controlled by a notebook personal computer (PC). The custom-made optical probe contained a 400-mm source fiber and a 200-mm detector fiber, with a source-detector separation of~350 mm.…”

“…Further detail of the optical setup has been described previously. 12,13,15 Twenty-nine unloaded tendons from 8 animals were investigated for backscatter anisotropy with probe positions at 08 and 1808 (parallel) and 908 and 2708 (perpendicular) to the long axis of the unloaded tendon. 12 Figure 1 shows the mean spectra from the second forepaw digit (F2) for perpendicular and parallel probe positions; there was no statistically significant difference ( p < 0.05) between different animals.…”

“…Images were then analysed by using NIH Image software as previously described (Marenzana et al 2002). The total surface area was then used to normalize peak force values and to obtain adhesive strength values.…”
