Optimization of chromosome doubling treatment for efficient in vivo doubled haploid production in maize

Abstract: An investigation to optimize the protocol for application of colchicine for enhancing the doubled haploid production in maize was done. 106 maize genotypes were used as maternal parents, whereas, pollen source involved tropically adopted haploid inducer (TAIL P1 and TAIL hybrid). After the elimination of chromosomes of inducer lines, haploid seeds were obtained from the crosses. Haploid seedlings were treated with three different doses, such as 0.04, 0.06 and 0.08 per cent of colchicines for different duration… Show more

“…Figure 3 shows the histograms of haploid and diploid plants. In the histogram of a diploid sample, two high G1 peaks of diploid cells can be observed, indicating that the plant presents a variation in the chromosome number in the same tissue (Kumar et al, 2021; Pires et al, 2019; Rádi et al, 2020). All detected ploidies were obtained by comparing the peak of the histograms of the duplicated plants with the peak of the histograms of the hybrid parents, and the external standard used was Pisum sativum .…”

“…Liguleless genotypes, along with molecular markers, are useful as testers for maintaining high HIR in haploid inducers. Doubled haploid technology is beginning to be adopted in Indian public maize breeding programmes as well (Khulbe et al, 2020;Kumar et al, 2021). Besides producing DH lines using the in vivo maternal haploid induction system, indigenous maternal haploid inducer lines F I G U R E 3 Normal and liguleless seedlings.…”

Besides producing DH lines using the in vivo maternal haploid induction system, indigenous maternal haploid inducer lines have been developed (Gain et al, 2022) to enhance access to DH germplasm.…”
