Optimized in vivo brain glutamate measurement using long‐echo‐time semi‐LASER at 7 T

Abstract: A short echo time (T ) is commonly used for brain glutamate measurement by H MRS to minimize drawbacks of long T such as signal modulation due to J evolution and T relaxation. However, J coupling causes the spectral patterns of glutamate to change with T , and the shortest achievable T may not produce the optimal glutamate measurement. The purpose of this study was to determine the optimal T for glutamate measurement at 7 T using semi-LASER (localization by adiabatic selective refocusing). Time-domain simulati… Show more

“…Most long‐TE studies did not report metabolites other than the singlets, especially in the hippocampus 45 . It was shown, however, that a long TE benefits Glu detection without MM contamination 46 . Based on T 2 relaxation times of macromolecules 21 and simulations of metabolites, we used a TE of 120 ms, at which the Glx multiplet is coalesced into a pseudo ‐singlet, quantifiable with high accuracy due to its prominent signal and the flatter or nonexistent MM background present at long TEs.…”

“…Our study has a number of strengths. We used 7T-MRS sequence with improved specificity to detect GSH resonance with reduce macromolecular interference [18,37]. Among the MRS studies specifically optimized for GSH detection, 7T studies [22,38] report higher effect sizes for GSH reduction in schizophrenia compared to 3T [12,39].…”

“…5). Water-subtracted spectra were modeled using the fitMAN, a-prior-knowledge based minimization algorithm, and a quantification template including 17 metabolite spectral signatures derived from simulation [52]. Our fitting template included 17 metabolites (alanine, aspartate, choline, creatine, GABA, glucose, glutamate, glutamine, GSH, glycine, lactate, myo-inositol, N-acetyl aspartate, N-acetyl aspartyl glutamate, phosphorylethanolamine, scyllo-inositol, and taurine).…”
