Optimized Transduction of Canine Paediatric CD34+ Cells Using an MSCV-based Bicistronic Vector

Abstract: We have used a murine MSCV-based bicistronic retroviral vector, containing the common gamma chain (gammac) and enhanced green fluorescent protein (EGFP) cDNAs, to optimize retroviral transduction of canine cells, including an adherent canine thymus fibroblast cell line, Cf2Th, as well as normal canine CD34(+) bone marrow (BM) cells. Both canine cell types were shown to express Ram-1 (the amphotropic retroviral receptor) mRNA. Supernatants containing infectious viruses were produced using both stable (PA317) an… Show more

“…Lentiviral infections of B-cells were performed using the centrifugation method 109 . In brief, 7 to 14×10 6 peripheral blood B cells were re-suspended in 5 to 10 ml of lenti-viral supernatant with 8 µg/mL final concentration of polybreme (Sigma-Aldrich).…”

“…In brief, 7 to 14×10 6 peripheral blood B cells were re-suspended in 5 to 10 ml of lenti-viral supernatant with 8 µg/mL final concentration of polybreme (Sigma-Aldrich). The mixture was centrifuged in 6 well plates for 1.5 h at 2,500g at 32°C 109 in order to achieve close contact between the retroviral particles and the cells. After 24 h, the media was replaced with fresh RPMI complete (at 1×10 6 cells/mL), and cells were cultured at 37°C and 5% CO 2 .…”

“…Cells were isolated as described elsewhere (27) and cultured in Dulbecco's modified Eagle's medium (DMEM) with L-glutamine, penicillin/streptomycin, and 10% fetal bovine serum (FBS). Spinfection was performed at 70% confluency with replication-deficient adenovirus expressing Cre recombinase to inactivate Smad3 in cultured chondrocytes from Smad3 fl/fl mice (28). As a negative control, cells were infected with adenovirus expressing green fluorescent protein (GFP).…”

“…Adenovirus with GFP/Cre-recombinase or GFP were added to the chondrocytes at 400 pfu/cell. Analysis of GFP expressing cells 12 h after infection verified that this protocol yields a 90% infection rate ( Suter et al, 2006 ). All experiments used cells at third passage (after harvest digestion, infection and appropriate media change) at day 2 after initial harvest.…”
