Dwarf surf clam, Mulinia lateralis, is widely considered as a model species for bivalves. The development of a standard culture system could greatly promote the production of high-quality individuals, yet information on the culture conditions for M. lateralis spat is still limited. This study aims to determine the suitable microalgae diet, temperature, and salinity for M. lateralis spat culture. The typically fed microalgae species, including Chlorella pyrenoidesa (Cp), Platymonas helgolandica (Ph), Dunaliella salina (Ds), Nitzschia closterium (Nc), and Chaetoceros muelleri (Cm), could be taken up by M. lateralis spat, and their filtration rates on Cp, Nc, and Cm were higher than those on Ds and Ph. For the entire spat culture, all diet trials showed similar survival percentages, while the mono-specific diet Cp exhibited the highest growth rate, suggesting that Cp was the optimal microalgae species for M. lateralis spat. Through simultaneously maximizing the growth and survival of spat, the optimal microalgae concentration and stocking density were 5 × 104 cells ml–1 and 400–600 individuals m–2 for 30–40-day-old spat as well as 1 × 105 cells ml–1 and 400–600 individuals m–2 for 40–50-day-old spat, respectively. In addition, the spat had higher growth rates and survival percentages at the temperature of 20–22°C and salinity of 22–25 ppt. The results of this study provide a basis for further culture of M. lateralis spat, and the optimized conditions will be of great significance for the construction of the standard culture system of M. lateralis.

This study investigated filtration and ingestion rates of various microalgae species by hard clam, Mercenaria mercenaria, larvae (veliger, pediveliger) and post-set. Four microalgae species, Tisochrysis lutea, Pavlova lutheri, Chaetoceros gracilis and C. muelleri were evaluated for larvae, and an additional species Cyclotella nana, were evaluated for post-set. Flagellates (T. lutea and P. lutheri) were filtered and ingested at higher rates than diatoms (C. gracilis, C. muelleri and C. nana) by both larvae and post-set.T. lutea was filtered and ingested at a higher rate than all other microalgae species evaluated. The filtration rate of T. lutea was 11.2 µl/ hr, 26.6 µl/ hr and 342.2 µl/ hr by veliger larvae, pediveliger larvae and post-set respectively. Ingestion rate of T. lutea was 369 cells/hr, 1441 cells/hr and 10,357 cells/hr by veliger larvae, pediveliger larvae and post-set respectively. In contrast, C. muelleri was filtered and ingested at lower rates by larvae, and C. nana was filtered and ingested at lower rates by post-set compared with other microalgae species. The differences in filtration and ingestion rates among microalgae species were primarily attributed to the size, shape and dry weight of microalgae species.

Choice of microalgae diet affects production outcomes in bivalve hatcheries. This study aimed to identify an optimal microalgae diet to improve hard clam, Mercenaria mercenaria, juvenile production. Four microalgae species (Tisochrysis lutea, Pavlova lutheri, Chaetoceros gracilis and Cyclotella nana) in six dietary combinations were fed for 6 weeks; then, growth, survival and fatty acid profiles of juveniles were evaluated. The highest numerical growth and survival of juveniles were attained by feeding a tetra-species microalgae diet, but no statistical difference was found by feeding bior tri-species diets that consisted of a mix of flagellate and diatom microalgae. Significant differences in fatty acid profiles were found among clams fed different microalgae diets. A contrasting polyunsaturated fatty acid (PUFA) profile was found in juveniles fed flagellate-or diatom-only diets. Clams fed flagellate-only diets contained a higher percentage of docosahexaenoic acid (DHA) but a lower percentage of (n-6) PUFAs, whereas those fed diatom-only diets had a higher percentage of arachidonic acid (ARA) and eicosapentaenoic acid (EPA) but a lower percentage of DHA. In contrast, the percentage of EPA, DHA and ARA was neither very high nor very low in juveniles fed the tetra-species diet. The complementary nutritional profile of the tetra-species microalgae diet resulted in enhanced production and exhibited a more balanced fatty acid profile in clam tissue. The tetra-species microalgae diet should be a preferable choice for feeding hard clam juveniles, but the cost of producing multiple microalgae species for hatchery/nursery operations should be taken into consideration as bi-and trispecies diets provided statistically similar growth and survival. Results of this study can be used to improve productivity and profitability in M. mercenaria hatcheries.
