Orchestrating Lymphocyte Polarity in Cognate Immune Cell–Cell Interactions

Abstract: The immune synapse (IS) is a specialized structure established between different immune cells that fulfills several functions, including a role as a communication bridge. This intimate contact between a T cell and an antigen-presenting cell promotes the proliferation and differentiation of lymphocytes involved in the contact. T-cell activation requires the specific triggering of the T-cell receptor (TCR), which promotes the activation of different signaling pathways inducing the polarization of the T cell. Dur… Show more

“…APCs are able to present antigens to cognate naïve CD4+ and CD8+ T cells [ 185 ]. Upon antigen recognition, CD4+ T cells activate, proliferate, and differentiate into specialized effector T helper (Th) cells, whereas CD8+ T cells activate, proliferate, and differentiate into CD8+ cytotoxic T lymphocytes (CTL) [ 186 , 187 ].…”

“…T cell activation also necessitates the movement of the microtubule organizing center (MTOC) towards the T cell-APC contact site. The relocated MTOC then guides the polarization of intracellular vesicles to the IS [ 185 ].…”

“…The orchestration of signaling by surface receptors as well as signaling molecules that accumulate at the IS area crucially depends on the polarization of the microtubule-organizing center (MTOC) 19 , which brings the endosomal recycling compartment at the T cell:APC contact region, thus favoring directional intracellular trafficking 20 . In naïve T cells, a number of molecules, including receptors (for example, the TCR) and membrane-associated regulators—for example, the lymphocyte-specific protein tyrosine kinase (Lck) and the linker for activation of T cells (LAT)—are clustered to the synaptic area not only from plasma membrane-associated pools but also from intracellular endosomal pools 2 .…”

“…LAT associates with SLP76 that recruits multimolecular complexes. PLCc1 being one of the multimolecular complexes ligates with LAT and induces activation of PLCc1Á Activated PLCc1 then reacts with phosphatidylinositol-4,5-bisphosphate at the plasma membrane generating diacylglycerol (DAG) and inositol-1,4,5-trisphosphate (IP3) (Bustos-Mor an et al 2016). This IP3 then ligates with the IP3 receptor (IP3R) on endoplasmic reticulum (ER), thereby releasing Ca 2+ from ER.…”
