Ordered recruitment of histone acetyltransferases and the TRAP/Mediator complex to thyroid hormone-responsive promoters in vivo

Abstract: Transcriptional coactivators implicated in gene activation by the thyroid hormone receptor (TR) include members of the p160͞ steroid receptor coactivator (SRC) family of proteins, p300, and the multisubunit TR-associated protein (TRAP)͞Mediator complex. We investigated the temporal recruitment of these cofactors to mammalian thyroid hormone (T3)-responsive promoters in vivo. We show that upon T3 treatment, TR recruits all three types of coactivators to specific promoters in at least two sequential steps: p160͞… Show more

“…This result was consistent with previous reports that indicated a requirement for p53 in the downregulation of FoxM1 (26,35). The FoxM1 mRNA expression levels substantially decreased when E2F1 was knocked down in MCF7 cells; this indicated a possible involvement of E2F1 in FoxM1 transcription (35).…”

“…Each lysate was incubated with 1 mg of anti-AhR (Biomol), antiRelA (C-20, Santa Cruz Biotechnology) or anti-p50 (Cell Signaling) antibodies and PureProteome Protein A Magnetic Bead (Millipore) for chromatin immunoprecipitation assay. Chromatin immunoprecipitation analysis was performed using a procedure with the following modifications (Sharma and Fondell, 2002). The chromatin immunoprecipitation and input DNA were used for the real-time PCR analysis.…”

“…In the last few years various investigators have shown that coactivator complexes including p160/SRC and DRIP are recruited to steroid hormone-regulated genes by nuclear receptor in a sequential and mutually exclusive manner [14][15][16][17]. The ordered association of transcriptional regulators exhibits binding kinetics with periods of 40 to 60 minutes.…”
