Osteopontin: regulation in tumor metastasis

Abstract: Osteopontin is a secreted phosphoprotein that has been implicated as an important mediator of tumor metastasis and has been investigated for use as a biomarker for advanced disease and as a potential therapeutic target in the regulation of cancer metastasis. The OPN DNA sequence is highly conserved and the protein contains several important functional domains including alpha(v)beta integrin and CD44 binding sites. High levels of OPN expression correlate with tumor invasion, progression or metastasis in multipl… Show more

“…Elevated OPN expression is associated with advanced stage, tumor invasion, and metastasis in multiple cancers. The function of OPN is versatile and involves the induction of multiple signalling pathways (PI 3'-kinase/Akt, NF-kB) by integrin and CD44 receptors that mediate metastatic processes [8,18,19].…”

“…Osteopontin (OPN) is a multifunctional extracellular matrix protein that is produced by numerous cell types [6][7][8][9][10][11]. OPN plays important roles in cancer cell adhesion, cell motility, and survival [12][13][14][15][16].…”

“…Les SIBLING pourraient jouer un rôle capital dans la formation des métastases car elles sont impliquées, comme décrit précédemment, dans l'adhésion cellulaire, la migration et la dégrada-tion de la matrice. Il a été démontré dans plusieurs études que l'OPN et la BSP jouent un rôle prométastatique (pour revue, voir [30]). Les tumeurs susceptibles d'évoluer vers un stade plus avancé présentent une expression de SIBLING augmentée ou apparaissant de novo.…”

“…These proteins are important players in cell-cell and cell-matrix communications. 7 Human OPN has a size of 314 amino acids, [8][9][10] but is detected in bands with apparent sizes of 41-75 kDa. This is due to different splice variants and intensive post-translational modifications.…”

“…[17][18][19][20][21][22][23] Interaction of OPN with avb3 integrin also alters the host immunity by increasing cytokine expression 24,25 and enhances angiogenic functions through avb3-integrinmediated activation of endothelial cells. 7,[26][27][28] In view of these important tumor progressive functions, Opn represents a potential therapeutic target for the modulation of tumor progression and metastasis formation. Therefore, it was the aim of this study to silence Opn expression by an antisense-oligomer in CC531 lacZ rat colorectal cancer cells.…”
