Osteoprotegerin exposure at different stages of osteoclastogenesis differentially affects osteoclast formation and function

Abstract: This study aimed to investigate the effects of osteoprotegerin (OPG), a decoy receptor for receptor activator for nuclear factor jB ligand (RANKL), during the various stages of osteoclast differentiation, and additionally investigate its effects on osteoclast adhesion and activity. RAW264.7 murine monocytic cells were incubated with macrophage colony-stimulating factor and RANKL for 1, 3, 5, or 7 days, followed by an additional 24-h incubation in the presence or absence of OPG (80 ng/mL). We examined osteoclas… Show more

“…We found that OPG could inhibit the bone resorption activity of OCs, characterized as diminished lacuna areas and decreased protein levels of Ctsk and TRAP, which agreed with previous observations (Zhao, Gu et al, ). Classical autophagy inducers, such as metformin (Buzzai et al, ), everolimus (Kneissel et al, ), and resveratrol (Morselli et al, ) were found to be beneficial to bone health in rescuing OVX‐induced bone loss, by inhibiting OC differentiation or bone resorption activity (Pierrefite‐Carle, Santucci‐Darmanin, Breuil, Camuzard, & Carle, ).…”

“…Furthermore, the effect of RANKL treatment on osteoclast differentiation and maturation was explored using a TRAP assay. TRAP is an early enzyme marker of osteoclastogenesis, which is readily detectable in mononuclear pre-osteoclasts, through to mature osteoclasts ( 18 ). Fig.…”

“…Osteoclastogenesis involves several stages, starting from the precursor cells, which differentiate into mature osteoclasts [ 59 ]. Knowledge on the regulation of osteoclast differentiation is important in providing insights on suitable therapeutic targets for intervening bone loss diseases like osteoporosis.…”
