“…Substantial data support actions of estrogen through the mesolimbic dopamine system of female rats (Sakamoto et al, 1993;Thompson and Moss, 1994;Bosse et al, 1997;Becker, 1999;Febo et al, 2003. Our present data support effects of estrogen in other brain regions as well.…”

“…Estradiol administration has been shown to increase dopamine receptor density over a period of days (Hruska 1986). Adult ovariectomy has also been shown to reduce the density of the dopamine transporter (DAT) in the nucleus accumbens and in the striatum (Bosse et al 1997), a region where DAT density is reduced in males compared to females (Rivest et al 1995). Further, cocaine-induced behavioral activity varies with alterations in estrogen levels across the estrus cycle in adult females: cocaine responses are blunted during diestrus compared to the proestrus and estrus phases (Becker and Cha 1989;Quinones-Jenab 1999;Walker et al 2002).…”

“…However, we found no alterations in the expression of genes encoding tyrosine hydroxylase, GTP cyclohydrolase 1, dopa decarboxylase, monoamine oxidase A and B, catechol-O-methyl-transferase. From these findings we conclude that the transmission of reward signals to the PFC, in particular its dopaminergic component, is largely dependent on the gonadal hormone milieu, and E2 regulates dopamine availability mainly through the modulation of intracellular signaling pathways [55,58]. The tuning of dopamine levels has particular importance since dopamine effects follow a U-shaped dose-response curve in the PFC [57,59,60].…”

“…A previous study has investigated estrogenic regulation of dopamine transporter mRNA expression in the VTA of OVX rats using in situ hybridization [58]. They have found hormonal regulation of dopamine transporter expression neither in OVX nor E2-replaced animals.…”
